چکیده

We tested the hypothesis that variants in the gene encoding the prepropeptide YY (PYY) associate with type 2 diabetes and/or obesity. Mutation analyses of DNA from 84 patients with obesity and familial type 2 diabetes identified two polymorphisms, IVS3 68C>T and Arg72Thr, and one rare variant, 151C>A of PYY. The common allele of the Arg72Thr variant associated with type 2 diabetes with an allele frequency of the Arg allele of 0.667 (95% CI 0.658–0.677) among 4,639 glucose-tolerant subjects and 0.692 (0.674–0.710) among 1,326 patients with type 2 diabetes (P 0.005, odds ratio 1.19 [95% CI 1.05–1.35]). The same polymorphism associated with overweight (25 < BMI < 30 kg/m) (P 0.018, 1.15 [1.02– 1.28]). In quantitative trait analyses of a population-based sample of 6,022 subjects, the Arg allele was associated with an increased plasma glucose level 2 h after an oral glucose tolerance test (OGTT) (P 0.03), an increased area under the curve for the post-OGTT plasma glucose level (P 0.03), and a lower insulinogenic index (P 0.01). In conclusion, the common Arg allele of the PYY Arg72Thr variant modestly associates with type 2 diabetes and with type 2 diabetes–related quantitative traits. Diabetes 54:2261–2265, 2005

Association of CpG-SNP and 3\'UTR-SNP of WFS1 with the Risk of Type 2 Diabetes Mellitus in an Iranian Population

Type 2 diabetes mellitus (T2DM) is one of the most common multifactorial disorders in Iran. Recent genome wide association studies (GWASs), and functional studies have suggested that WFS1 may predispose individuals to T2DM. However, to date, the possible association of such variants with T2DM in Iranians remained unknown. Here, we investigated the association of the two polymorphisms of WFS1 (r...

MAP3K1 May be a Promising Susceptibility Gene for Type 2 Diabetes Mellitus in an Iranian Population

Considering that MAPK (mitogen- activated protein kinase) signaling pathway has an important role in the progression of inflammatory cytokine secretion in type 2 diabetes mellitus (T2DM), we have recently investigated the reported genetic polymorphism from genome wide association study in MAP3K1 (mitogen-activated protein kinase kinase kinase 1) in diabetes as an important member of MAPK signal...

Genetic Polymorphisms of Estrogen Receptors in Iranian Women with Diabetes and Coronary Artery Disease

Estrogen might play an important role in the pathogenesis of diabetes mellitus type 2. Estrogens inhibit diabetes via distinct mechanisms particularly by reducing both hyperglycemia and plasma insulin levels. Estrogen exerts its physiological effects mainly through estrogen receptors including α and β types. Estrogen receptors are found in many tissues that participate in the pathogenesis of ty...

Association of two Common Single Nucleotide Polymorphisms (SNPs +45T/G and +276G/T) of ADIPOQ Gene with Coronary Artery Disease in Type 2 Diabetic Patients

Background: Adiponectin, an adipocyte-secreted hormone, is known to have anti-atherogenic, anti-inflammatory, and anti-diabetic properties. In the present study, the association between two common single nucleotide polymorphisms (SNPs) (+45T/G and +276G/T) of ADIOPQ gene and coronary artery disease (CAD) was assessed in the subjects with type 2 diabetes (T2DM). Methods: Genotypes of two SNPs we...

Birth Weight and Childhood Onset Type 1 Diabetes: A Case-Control Study in Shiraz, South of Iran

To determine the relation between birth weight and age of presentation of type 1 diabetes and assess its risk factors, a case-control study was done on all diabetic patients under the age of 15, who were registered in Shiraz Diabetic Center or admitted in Namazi Hospital. A case group of 87 diabetic patients were matched with a control group of 100 healthy individuals. Birth weight ≥ 4kg was de...
